1. Keep in mind that it is no load (make sure that the ultrasonic amplitude converter can be started after pulling out the sample)
2. The depth of the amplitude transformer (ultrasonic probe) into the water: about 1.5cm, the height of the liquid level should be more than 30mm, the probe should be centered, not attached to the wall. Ultrasonic wave is a vertical longitudinal wave, which is not easy to form convection if inserted too deep, affecting the crushing efficiency.
3, Ultrasonic processor parameter setting: set key good instrument task parameters (see the detailed setting book or call for consultation), about the temperature requirements of sensitive samples (such as bacteria) generally inside the ice bath, the practice temperature must be less than 25 degrees, protein nucleic acid will certainly not denaturation.
1) Time: the ultrasonic time should not exceed 5 seconds each time, and the interval time should be greater than or equal to the ultrasonic time to facilitate heat distribution. The time setting shall be short ultrasonic time, multiple ultrasonic times, extended ultrasonic machine and probe life.
2) Ultrasonic power: should not be too large to avoid sample spatter or foaming. If the sample capacity is less than 10ml, the power should be less than 200w. Use 2mm ultrasonic probe. The power of 10-200ml sample capacity is 200-400w. 6mm ultrasonic probe is selected and the amplitude transformer behind the panel is hit to the corresponding gear. The sample capacity power above 200ml is between 300-600w, 10mm ultrasonic probe is selected, and the amplitude transformer behind the panel is hit to the corresponding block; (power of small probe with 2MM is forbidden to exceed 350W)
3) Container selection: choose the beaker as many samples as possible, which is also beneficial to the convection of samples in ultrasound and the improvement of crushing efficiency.
For example; The best way to handle 20ML is to use 20ML beakers. For example, setting parameters of 100ml escherichia coli sample: ultrasonic 5 seconds/interval 5 seconds 70 times (total time is 10 minutes). The power of 300W (for reference only) is about 500ML, and the power is about 500w-800w
4. If the sample is placed in a 1.5ml EP tube, please make sure that the EP tube is fixed in order to avoid no-load caused by the drop of liquid level after ice bath ablation.
