The Conditions For Ultrasonic Processing of Streptomyces

1. Bacteria culture medium was centrifuged at 5000 RPM for 24 hours for 5 minutes to collect cells.
2. Wash 3 times with na2hpo-nah2po buffer at pH7.5, then mix the bacteria and buffer into 1:3 bacterial suspension. Place in a large plastic 40 mL tube
3. Put the large plastic test tube into the ice bath and break it with ultrasonic wave (power 200 W, 1/2 "probe, crush for 30 s, intermittent for 30 s).
4. The broken liquid was centrifuged at a high speed of 12000r/min for 30 minutes to collect cell fragments and supernatant.

The ultrasonic sterilization process is basically the same as above, that is, the washing tank can also be washed once with pre-cooled normal saline or pH8 tris-hcl. In addition, the ultrasound dose varied greatly with the amount of samples and bacteria.
